Dan Trejo er sønn av Alice Rivera og Dan Trejo, og som ungdom var han rusmiddelavhengig og levde en kriminell livsstil. Han satt flere ganger i fengsel, blant annet i San Quentin State Prison, hvor han ble statsfengselets boksemester i lettvekt og weltervekt.[trenger referanse]
Trejo gjennomgikk et tolvtrinns avvenningsprogram, og på et møte traff han en innen filmbransjen som fikk han med i en mindre rolle i en film. Etter dette har han spilt i mange filmer sammen med flere av toppskuespillerne i Hollywood, som Johnny Depp, Al Pacino, Nicolas Cage, Charles Bronson, George Clooney, Edward Furlong, John Malkovich, Robert De Niro, Harrison Ford, Val Kilmer, Antonio Banderas og i serien The Flash.
